The regular size suits general marking on stock up to about twice its 2" width, while the large size is sized specifically for 2 × 4 lumber and adds a center slot for stud layout; choose the large size if you're framing or working directly with dimensional lumber.
The end faces are machined square to within 1/4°, so the line stays accurate when it's carried across to the adjacent face.
